蔬菜水果中霜霉威的测定

        霜霉威是一种具有局部内吸作用的低毒杀菌剂,属氨基甲酸酯类农药。其杀菌机制主要是抑制病菌细胞膜成分的磷脂和脂肪酸的生物合成,进而抑制菌丝生长、孢子囊的形成和萌发,广泛用于茄子、马铃薯、葡萄、草莓等蔬菜水果中绵疫病、霜霉病、猝倒病、晚疫病、黑胫病等病害防治。但霜霉威农药的过量喷洒可能会造成食品安全隐患,对人体健康造成危害。

 

  2022年《国家食品安全监督抽检实施细则》(简称国抽)与2021年相比,水果类项下葡萄的霜霉威和霜霉威盐酸盐检验项目中新增《NY/T 1379-2007 蔬菜中334种农药多残留的测定 气相色谱质谱法和液相色谱质谱法》。

 

  迪马科技参考《NY/T 1379-2007 蔬菜中334种农药多残留的测定 气相色谱质谱法和液相色谱质谱法》,建立了SPE-GC-MS法测定蔬菜水果中霜霉威,利用乙腈提取,ProElut Carb/NH2复合柱净化;检出限与标准方法一致,为7.0μg/kg,回收率符合标准要求。本方案具有提取方法简单、回收率高、净化效果优异、方法稳定性好等优点,可供大家参考。

2022国抽新增食品安全项目:蔬菜水果中霜霉威的测定

 

 

1、适用范围

本方案适用于茄子、葡萄中霜霉威的检测,检出限为7.0μg/kg。

2、标准品配制

 

(1) 霜霉威标准储备液1000μg/mL:准确称取10mg标准品于10mL容量瓶中,用丙酮溶解并稀释至刻度,配制成1000μg/mL的霜霉威标准储备液;

(2) 霜霉威标准工作液5.0μg/mL、1.0μg/mL:准确移取霜霉威标准储备液,用丙酮稀释为浓度5.0μg/mL、1.0μg/mL的霜霉威标准工作液。

 

3、提取

 

(1) 取15g样品于50mL塑料离心管中,准确加入30mL乙腈,涡旋振荡5min,加入萃取盐包(Cat.#: 64521s),振荡1min,6000 rpm离心2min,准确收集10mL上清液;

(2) 将上清液在45℃水浴下减压蒸至约1mL,加入10mL乙腈,再蒸至约1 mL,待净化。

 

4、净化

 

ProElut CARB/NH2 1000mg/6mL (Cat.#: 64105)

(1)活化:加入5mL乙腈-甲苯(3-1),弃去流出液;

(2)上样:将待净化液加入柱中,收集流出液;

(3)淋洗:加入10mL乙腈-甲苯(3-1)分次淋洗,收集流出液;

(4)洗脱:加入25mL乙腈-甲苯(3-1),合并(2)、(3)、(4)流出液;

(5)重新溶解:将洗脱液在45℃水浴下蒸至近干,用丙酮定容至2.5mL,供GC-MS分析。

 

5、分析条件

 

色谱柱:DM-5MS,30mx0.25mmx0.25μm(Cat.#: 8221)

进样口温度:280℃

升温程序:初始温度70℃,保持5min,以25℃/min升温至150℃,再以3℃/min升温至160℃,再以20℃/min升温至280℃,保持10min。

载气:氦气

流速:1.00mL/min

进样方式:不分流进样

进样量:1.0μL

离子源温度:230℃

接口温度:280℃

溶剂延迟:8min

电子轰击电离源(EI):选择离子监测模式(SIM),分组监测见表1
